Det har gått över 20 år sedan utomjordingarna anlände till jorden. Mänskligheten förberedde sig då på ett fientligt möte med överlägsen teknologi, men ingenting hände. Utomjordingarna blev istället kvar som flyktingar på jorden. Varelserna placerades i provisoriska hem i Sydafrikas District 9, allt medan jordens nationer debatterade deras fortsatta öde. Men nu har människornas tålamod tagit slut. Ansvaret för utomjordingarna har därför kontrakterats MNU (Mulit-National United), ett privat företag utan intresse i utomjordingarnas välfärd. MNU kommer att göra enorma vinster om de kan få utomjordingarnas vapen att fungera. Men än så länge har alla sådana försök misslyckats eftersom aktiveringen av vapnen kräver utomjordisk DNA. Spänningarna mellan människor och utomjordingar kulminerar när MNU-tjänstemän inleder omlokaliseringen av alla icke-mänskliga individer från District 9 till ett nytt läger. En av MNUs fältarbetare, Wikus van der Merwe (Sharlto Copley), utlöser ett utomjordiskt virus som börjar förändra hans DNA. Wikus blir snabbt den mest jagade mannen i världen, tillika den mest värdefulle, eftersom han bär på nyckeln som kan avslöja hemligheterna om utomjordingarnas teknologier. Utstött och utan vänner finns till slut bara en plats för honom att hålla sig gömd; i District 9.
